this detection. According to the recommendation of the International Federation of Clinical Chemistry and
Laboratory Medicine (IFCC), ethylidene-4-NP-G7 should be used in diagnostic tests for the determination of
α-amylase and pancreatic α-amylase.
EPS-G7 is 4,6-Ethylidene-4-nitrophenyl-1,4-α-D-maltoheptaoside, CAS 96597-16-9, molecular formula is C₅₀H₇₇NO₃₈, and molecular weight is 1300.1. This substrate is specifically designed for α-amylase activity measurement. The introduction of an ethylidene protecting group significantly enhances its specificity for α-amylase.
EPS-G7 is primarily used in α-amylase assay kits for clinical biochemistry. The core reaction principle is as follows: α-amylase in the sample hydrolyzes the α-1,4 glycosidic bonds of the substrate. Through a series of coupled enzymatic reactions (typically involving α-glucosidase), 4-nitrophenol (PNP) is ultimately released. By monitoring the rate of PNP production at 405 nm (kinetic method), the α-amylase activity in the sample can be accurately calculated.
