QYM/SURFTECH series of BCIP/NBT membrane/precipitation substrates feature low background, high stability, sensitivity, and minimal batch-to-batch variation. The series provides blue and purple signal systems, along with BCIP/NBT substrates dedicated for immunohistochemistry (IHC). BCIP/NBT One Component Membrane Substrate plus produces an dark purple signal and is suitable for immunohistochemistry (IHC), as well as applications such as Western blot and microarray.

Instruction:

QYM/SURFTECH BCIP/NBT One Component Membrane Substrate plus is ready-to-use and should be equilibrated to the reaction temperature before application.

Chromogenic Reaction Procedure (Western Blot):

1. Wash: After alkaline phosphatase (AP) conjugate incubation, wash the membrane 3 times with TBST washing buffer.

2. Chromogenic Reaction: Add 1 mL substrate and incubate at 20-37 C until bands appear dark purple.

3. Stop: Wash 2-3 times with 1.5 mL deionized water to terminate the reaction before background color develops.

4. Detection: Analyze bands by visual inspection or by imaging to quantify grayscale values.
