Instruction:
QYM/SURFTECH TMB Blue One Component Membrane Substrate is ready-to-use and should be equilibrated to the reaction temperature before use.
Chromogenic Reaction Procedure (Western Blot):
1. Wash: After HRP conjugate incubation, wash the membrane 3 times with PBST/TBST washing buffer;
2. Chromogenic Reaction: Add 1 mL substrate and incubate at 20-37 C until bands appear dark blue;
3. Stop: Stop the reaction by washing 2-3 times with 1.5 mL deionized water before background develops;
4. Detection: Analyze bands by visual inspection or by imaging to quantify grayscale values.
